Summary of Position

The Research Associate II / Associate Scientist I position will support preclinical development of MacroGenics antibody-drug conjugates (ADC) through cell line engineering and molecular biology support. This position will independently develop and engineer cell lines, and conduct cell-based, molecular, and biochemical assays which contribute to the ADC research and development pipeline.

Responsibilities and Job Duties

  • Develops stable cell lines using techniques that deliver and integrate DNA/RNA into the host genome using lentivirus transduction, lipid-based transfection, and electroporation methods to support preclinical ADC research and development.
  • Executes gene editing strategies to generate stable knockout cell lines using the CRISPR/Cas9 system and designs assays to monitor editing efficiency in support of target selection and preclinical research.
  • Maintains and characterizes cancer cell lines and establishes banks for experimental purposes using aseptic cell culture techniques supporting research within the Targeted Therapeutics Group.
  • Utilizes molecular biology techniques to conduct experiments involving DNA/RNA isolation and purification, expression construct design, molecular cloning and sub-cloning, RT-qPCR, PCR, sequencing, genotyping, and RNA profiling, contributing to preclinical and clinical research programs.
  • Designs and executes cell-based and biochemical assays to validate engineered cell lines and targets through flow cytometry, reporter assays, ELISA, western blotting, and cytotoxicity assays to select lead preclinical candidates.
  • Collects and analyzes data, interprets results, and presents results at internal research meetings contributing to go/no-go decisions on preclinical ADC programs.
  • Engages in problem solving and the identification and development of new methodologies to improve efficiencies, expand capabilities, and advance projects from research into development.
  • Executes a range of laboratory experiments efficiently and with high reproducibility to advance ADC research programs and overall corporate objectives.

Education & Other Credentials

  • Bachelor's degree in a scientific discipline with minimum of two years of related laboratory experience
  • Master's degree in a scientific discipline with related laboratory experience

Associate Scientist I

Education & Other Credentials

  • Bachelor's degree in a scientific discipline with minimum of five years of related laboratory experience
  • Master's degree in a scientific discipline with minimum of two years of related laboratory experience

Knowledge, Skills and Abilities

  • Experience in molecular biology and mammalian cell expression technologies.
  • Strong molecular biology background with a proficiency in DNA/RNA isolation and purification, cloning, sub-cloning, PCR, RT-qPCR, genotyping, expression construct design, transfection, CRISPR gene editing, and RNA profiling.
  • Proficient in engineering stable overexpressing and CRISPR gene knockout cell lines and performing molecular and cell-based assays to validate the engineered cell lines.
  • Hands-on experience with aseptic mammalian cell culture.
  • Ability to execute experiments, develop assays, optimize protocols, and interpret experimental results.
  • Capacity to manage multiple projects simultaneously and work effectively in a fast-paced, adaptive, collaborative environment.
  • Ability to work independently, with minimal supervision.
  • Problem solving skills, attention to detail, organized, ability to use good judgment.
  • Ability to keep detailed and accurate notebooks and computer records and maintain/update data in computer-based applications/databases.
  • Demonstrated strong written and oral communication skills, and ability to present results in a group setting

Preferred Qualifications

  • Experience with various software applications (Microsoft, GraphPad Prism, SnapGene, FloJo, BioIt, FreezerPro).
  • Biochemistry and protein chemistry experience (flow cytometry, FACS, ELISA, western blotting, SDS-PAGE, etc.)
